Expression and clinical significance of PBK/TOPK in monocytes ofpatients with rheumatoid arthritis

Abstract Objeetive 'To investigate the expression and clinical significance of PDZ-binding-kinase/T-LAK cell-.derived protein kinase ( PBK/TOPK) in monoeytes of patients with rheumatoid arthritis ( RA ). Methods PBK/TOPK expression in peripheral blood mononuclear cells ( PBMC) was detected by flow cytometry in 47 RA patientsand 20 healthy subjects. Results The expression level of PBK/TOPK in peripheral blood CD14* monocytes in RA patients was higher than that in healthy subjects, and the difference was statistically significant ( P<0.05 ). Theproportion of CD14* monoeytes in PBMC , white blood cell ( WBC) count, rheumatoid factor ( RF ) , immunoglobulinA ( lgA )and immunoglobulin G (lgG) of CD14* monoeytes in RA patients with low expression of PBK/TOPK werehigher than those in RA patients with high expression of PBK/TOPK. The difference was statistically significant( P<0.05 ). 'The monocyte PBK/TOPK expression in inpatients with RA was lower than that in outpatients, and the RFlevel in inpatients was higher than that in outpatients. The differenee was statistically significant ( P<0.05 ).Conclusion 'The low expression of PBK/'TOPK in monocytes is related to the inflammation of the disease, and it hascertain significance for the targeted therapy of RA.
